The Relationship Of EPO Over-Expression To VHL Mutations And Hif1α And 2α In SCCRCC

ORLANDO, FL (UroToday.com) - Pathogenesis of clear cell renal cell carcinoma (RCC) involves a mutation in the VHL gene, resulting in upregulation of hypoxia-inducible factor (HIF).

The authors evaluated expression of erythropoietin (EPO), a target gene of HIF, and correlated this to expression of HIF-1alpha and HIF-2alpha in 82 patients with sporadic RCC. EPO expression was found in 65% of tumors that expressed HIF-1alpha and in 84.1% of tumors that were HIF-2alpha positive.

Targeted disruption of the HIF pathway has proven successful in medical treatment of advanced RCC, and further delineation of the HIF pathway through investigations such as this are necessary to catalyze intelligent drug design.
